    Predictors of continued HIV-risk behaviors among drug users in methadone maintenance therapy program in China—A prospective study

    BACKGROUND: To examine the predictors of continued drug- and sex-related HIV-risk behaviors among drug users in methadone maintenance therapy (MMT) programs in China. METHODS: We followed a sample of 5,035 drug users enrolled for the first time in MMT programs at baseline, 6 months, and 12 months utilizing a longitudinal prospective study design. Drug users’ HIV-risk behaviors, MMT characteristics, and drug use, were assessed at all three waves using a structured interview and HIV/HCV status was assessed at baseline and 12-month follow-up using biological specimens. RESULTS: The point prevalence of HIV was 7.6% and 78.4% for HCV at baseline. Results of generalized linear mixed logistic regression models revealed that HIV-positive MMT clients were more likely to engage in drug injection (aOR = 1.70) and syringe sharing (aOR = 4.73). HCV-positive clients were more likely to inject drugs (aOR = 2.58), share syringes (aOR = 1.97), and have multiple sexual partners (aOR = 1.47). Adherence to MMT was the most significant predictor of reduced HIV-risk behaviors. CONCLUSIONS: Our data confirmed the positive effects of MMT on HIV prevention and underscored the urgency for programs to reduce HIV risk in HIV- and HCV-positive clients. There is a pressing need to strengthen existing counseling services for HIV-positive drug users to reduce their drug-related risk behaviors and to provide counseling for HCV-positive drug users. Further studies are needed to explore interventions to address high dropout rates and low adherence among MMT clients

    Preparation and evaluation of tamsulosin hydrochloride sustained-release pellets modified by two-layered membrane techniques

    AbstractThe aim of the present study was to develop tamsulosin hydrochloride sustained-release pellets using two-layered membrane techniques. Centrifugal granulator and fluidized-bed coater were employed to prepare drug-loaded pellets and to employ two-layered membrane coating respectively. The prepared pellets were evaluated for physicochemical characterization, subjected to differential scanning calorimetry (DSC) and in vitro release of different pH. Different release models and scanning electron microscopy (SEM) were utilized to analyze the release mechanism of Harnual® and home-made pellets. By comparing the dissolution profiles, the ratio and coating weight gain of Eudragit® NE30D and Eudragit® L30D55 which constitute the inside membrane were identified as 18:1 and 10%–11%. The coating amount of outside membrane containing Eudragit® L30D55 was determined to be 0.8%. The similarity factors (f2) of home-made capsule and commercially available product (Harnual®) were above 50 in different dissolution media. DSC studies confirmed that drug and excipients had good compatibility and SEM photographs showed the similarities and differences of coating surface between Harnual® and self-made pellets before and after dissolution.     Cloning of a gene encoding glycosyltransferase from Pueraria lobata (Wild.) Ohwi and its expression in Pichia pastoris

    The key enzyme of puerarin biosynthesis in Pueraria lobata (Willd.) Ohwi was unclear but may involve glycosylation. To investigate the regulation of puerarin biosynthesis, a putative UDP-dependent glycosyltransferase (UGT) gene, PlUGT1 was isolated from P. lobata root, which contained abundant puerarin. PlUGT1 encoded 480 deduced amino acid residues with a conserved UDP-glucose-binding domain, which has 61 to 84% similarity to homologues from other plant species. SDS polyacrylamide gel electrophoresis and western blotting results showed that, fusion protein migrated as a single protein band with a molecular weight of 55 kDa. A yeast expression vector pPICZA-PlUGT1 was constructed and was transformed into Pichia pastoris strain GS115. Several recombinants containing multi-copy expression cassettes were obtained on the zeocin-YPD plate and confirmed by southern dot blotting. The yield of PlUGT1 attained 0.05 g/l when recombinant cells were cultured at pH 5.5, 30°C and induced with 0.5% methanol for 72 h. The expression of PlUGT1 protein correlates positively with the copy numbers of PlUGT1 in transformed yeast cells. These results suggest that, the PlUGT1 protein can be expressed efficiently in the P. pastoris expression system and may supply a new economic and convenient way for the production of PlUGT1 protein.Keywords: Pueraria lobata (Willd.)     Silencing SARS-CoV Spike protein expression in cultured cells by RNA interference

    AbstractThe severe acute respiratory syndrome (SARS) has been one of the most epidemic diseases threatening human health all over the world. Based on clinical studies, SARS-CoV (the SARS-associated coronavirus), a novel coronavirus, is reported as the pathogen responsible for the disease. To date, no effective and specific therapeutic method can be used to treat patients suffering from SARS-CoV infection. RNA interference (RNAi) is a process by which the introduced small interfering RNA (siRNA) could cause the degradation of mRNA with identical sequence specificity. The RNAi methodology has been used as a tool to silence genes in cultured cells and in animals. Recently, this technique was employed in anti-virus infections in human immunodeficiency virus and hepatitis C/B virus. In this study, RNAi technology has been applied to explore the possibility for prevention of SARS-CoV infection. We constructed specific siRNAs targeting the S gene in SARS-CoV. We demonstrated that the siRNAs could effectively and specifically inhibit gene expression of Spike protein in SARS-CoV-infected cells.     Detecting and pyramiding target QTL for plant- and grain-related traits via chromosomal segment substitution line of rice

    IntroductionPlant height and grain length are important agronomic traits in rice, exhibiting a strong effect on plant architecture and grain quality of rice varieties.MethodsMethods: A novel rice chromosomal segment substitution line (CSSL), i.e., CSSL-Z1357, with significantly increased plant height (PH) and grain length (GL) was identified from CSSLs constructed by using Nipponbare as a receptor and a restorer line Xihui 18 as a donor. Seven agronomic traits of PH, PL, GL, GW, GPP, SPP, and TGW were phenotyped, and REML implemented in HPMIXED of SAS were used to detect the QTL for these traits. Secondary CSSLs were screened out via marker-assisted selection (MAS) to estimate the additive and epistatic effects of detected QTLs, evaluating the potential utilization of pyramiding the target QTLs for yield and quality improvement of rice varieties.Results and DiscussionResults and Discussion: CSSL-Z1357 carried nine segments from Xihui 18 with an average segment length of 4.13 Mb. The results show that the long grain of CSSL-Z1357 was caused by the increased number of surface cells and the length of the inner glume. Thirteen quantitative trait loci were identified via the F2 population of Nipponbare/CSSL-Z1357, including three each for GL (qGL-3, qGL-6, and qGL-7) and PH (qPH-1, qPH-7, and qPH-12I), among which qGL-3 increased GL by 0.23 mm with synergistic allele from CSSL-Z1357. Additionally, three single (S1 to S3), two double (D1, D2), and one triple segment (T1) substitution lines were developed in F3 via MAS. Results show that pyramiding the segments from Chr.3 (qGL-3 and qPH-3), Chr.6 (qGL-6 and qPH-6), and Chr.7 (Null and qPH-7) tended to result in better phenotype of increased GL and PH and decreased grain width, providing a potential basis for enhancing grain yield and quality in rice breeding

    BACKGROUND(#br)Schizochytrium has been widely used in industry for synthesizing polyunsaturated fatty acids (PUFAs), especially docosahexaenoic acid (DHA). However, unclear biosynthesis pathway of PUFAs inhibits further production of the Schizochytrium. Unsaponifiable matter (UM) from mevalonate pathway is crucial to cell growth and intracellular metabolism in all higher eukaryotes and microalgae. Therefore, regulation of UM biosynthesis in Schizochytrium may have important effects on fatty acids synthesis. Moreover, it is well known that UMs, such as squalene and β-carotene, are of great commercial value. Thus, regulating UM biosynthesis may also allow for an increased valuation of Schizochytrium.(#br)RESULTS(#br)To investigate the correlation of UM biosynthesis with fatty acids accumulation in Schizochytrium, fluconazole was used to block the sterols pathway. The addition of 60 mg/L fluconazole at 48 h increased the total lipids (TLs) at 96 h by 16% without affecting cell growth, which was accompanied by remarkable changes in UMs and NADPH. Cholesterol content was reduced by 8%, and the squalene content improved by 45% at 72 h, which demonstrated fluconazole’s role in inhibiting squalene flow to cholesterol. As another typical UM with antioxidant capacity, the β-carotene production was increased by 53% at 96 h. The increase of squalene and β-carotene could boost intracellular oxidation resistance to protect fatty acids from oxidation. The NADPH was found to be 33% higher than that of the control at 96 h, which meant that the cells had more reducing power for fatty acid synthesis. Metabolic analysis further confirmed that regulation of sterols was closely related to glucose absorption, pigment biosynthesis and fatty acid production in Schizochytrium.(#br)CONCLUSION(#br)This work first reported the effect of UM biosynthesis on fatty acid accumulation in Schizochytrium. The UM was found to affect fatty acid biosynthesis by changing cell membrane function, intracellular antioxidation and reducing power. We believe that this work provides valuable insights in improving PUFA and other valuable matters in microalgae

    Fiscal decentralization and horizontal fiscal inequality in China: New evidence from metropolitan areas

    If fiscal decentralization reform and unbalanced economic growth increase horizontal fiscal inequality across China, this may deteriorate public services in underfunded regions and slow down economic growth and urbanization in the long run. Our study investigated the inequality of fiscal expenditures in metropolitan areas in China during the period 1999-2008. Although fiscal inequality increased before 2001, we found that a decreasing pattern was evident afterward. We also found that while revenue inequality was highly correlated with observed disparities in expenditures, inequality of transfers from central and provincial governments also contributed, although to a smaller extent. Guangdong, one of the most developed provinces in China, surprisingly had the highest level of intraprovincial fiscal inequality

    Preparation and evaluation of taste masked oral suspension of arbidol hydrochloride

    The purpose of this study was to cover the bitter taste of arbidol hydrochloride (ARB) and develop dry suspension with combination of solid dispersion and flavors. Taste masking was successfully done by solid dispersion using octadecanol as the carrier by fusion method. Suspending agents, carriers and other excipients were selected. Differential scanning calorimetry (DSC) and Fourier transform infrared spectroscopy (FTIR) were performed to identify the physicochemical interaction between drug and carrier, DSC analysis indicated that ARB was amorphous in the solid dispersion, FTIR spectroscopy showed no interaction between drug and carrier. Taste masking was evaluated on six volunteers with a score of 4.9. The results demonstrated successful taste masking. Water was used to study the in vitro dissolution performance of the three formulations of commercial tablet, capsule and self-made suspension. The self-made suspension showed a lower and slower release, the insoluble carrier octadecanol blocked the drug dissolving from the solid dispersion. It was indicated from the primary stability study, the self-made suspensions were sensitive to high temperature, high humidity and strong light conditions, they should be stored in sealed containers away from heat, light and humidity
